Krogan “Poison Ivy” Gladiator
Annihilation: 4a/5b/6b 4a = radius over recharge penalty as annihilation is your prime primer (wordplay) and you will use the other two skills less frequently anyway 5b = movement speed better than additional damage from annihilation; ever wanted to see a Krogan running on speed? Here you go! (Still slower than Asari Expert and Angara Avenger of course, but definitely quick!) 6b = simple and easy – let them fly (the unarmored/unshielded cannon fodder) cause it leaves a certain amount of foes helpless for some time allowing to take them out via combo detonations and/or weapons by yourself or your team mates; veeery helpful and nice; at least one! opportunity to fight back adhi and wraith.

Pull: 1 point only as I prefer to Nova and Pull doesn’t work on armored enemies anyway. BUT, should you prefer Pull, then I would go for 4b (radius)/5a (damage while pulled)/6a (killing shields to prime them for next pull)
Nova: 4a/5a/6a 4a = damage over radius as 4m radius already is a lot and I prefer to kill annihilation-primed foes by one Nova then priming larger radius (which rarely works anyway) 5a = armor over shield damage 6a = damage, damage, damage; but less due to the additional damage but more to avoid the shield-driven Nova which takes out 100% of your shields leaving you totally unshielded for a few seconds which is no good advice imho; the complete replenishment of your shields takes about the same time as recharging your power costs so no benefit anyway; with a power efficiency booster you rather run and nova with full shields instead of depleting your shields fully with each nova waiting for already 3-4 seconds before Nova-ing and depleting shields again.

Offensive biotics: 4a/5b/6a 4a = damage over power recharge (enhances Nova/Annihilation) 5b = enhances weapons and weight capacity allowing you to carry even the heaviest weapon combinations – cool! 6a = biotic damage/force over melee/weapon as weapons are strong enough anyway and melee you won’t use as often (unprecise, takes too long and can be easily interrupted by attacking foes)
Rage: 4a/5a/6b 4a = health and shields over melee, rationale already mentioned 5a = health and shield regeneration over melee, I’m repeating myself… 6b = forget melee and take health and shields…
Health: 1141 / Shield: 530 with this build, the strongest kit in the game so far, can take a lot of damage which helps running around with annihilation field and camping in hack/upload zones help keeping these free of adhi/wraith/breacher (the last one at least partially) plus can carry the REALLY heavy guns.

The build is based on a rank X level 20 character. Obviously you won't start with a rank X card hence you won't be able to distribute all 85 skill points from the very beginning even if leveled up to 20 already which is even more highly recommended. If so my recommendation is to go for annihilation first, then Nova or Pull based on your preference.
Equipment: Adaptive War Amp is the one to go as it will even more strengthen your annihilation field and pull and/or nova. Juggernaut shield again could be taken but only if you are going to melee as additional shields are not necessarily required.
Booster: 1st choice is Power Efficiency to get pull/nova going more often, that’s it. 2nd one depending on your preference and map/faction.

What makes her special? The Gladiator is a hybrid from an expert (Annihilation/Pull) and a vanguard (Nova) with heavy weight on the expert skills. Among the "weak" Krogans definitely the strongest and most durable. Also she can take a hell of heavy weapons.
Her Krogan hammer melee attack is also special and might be good however personally I struggle with the load time and the lack of aim. I never know whether to aim for the target or sligthly left of it, most often I try the latter. Anyway as your melee easily can be interrupted by foes (adhis, berserkers especially) it's not my first choice to melee with her. Not that it's not possible at all but Annihilation plus Nova plus weapon simply works better/quicker/more reliable for me. (Edit: after the 1.08 patch’s melee buff now at least there is some noticeable damage with her Krogan hammer. Aim is still terrible…)

What is unique about her? First of all combining the Asari expert's annihilation with a fast moving Krogan tank is hell of fun. Running around, priming targets, Nova, shoot lower level enemies up in the air? A lot of fun.
Second she can absorb a lot of enemy aggro saving your team mates plus can swallow damage while reviving or finishing objectives. I wouldn't consider her my first choice for soloing but she can be a strong member in any gold mission priming enemies for team mates' detonations and helping cleaning out the enemy crowd.
Basically the Asari expert Krogan tank version just slower/stronger and clearly a movable heavy weapon platform. Soned, Piranha, Widow, Black Widow would be a few of well feasible weapons and among my choices. She also is a strong asset when clearing hack and upload circles from adhis/wraiths and breachers. As always - Annihilation permanently on as it delivers constant damage plus higher movement speed with above build. Although that's never my goal it's easy to top score in a lobby or at least not being the last one. But your score doesn't really matter. The team counts.
Have fun.
