It feels a bit weird to say, but I'm technically on my first playthrough, about to start Trespasser.
It's a playthrough from 2015 before the DLC existed. Since reloading it, I've done Jaws and Descent. I believe I have all the rune recipes, and any schematics that would be considered "important." Other than requisitions, the journal is empty of quests (except for Bottles on the Wall, but I'm googling that one because I have one left to find.)
I figured I'd ask if anyone can think of anything I'd want to make sure I have, first. I didn't know about Sulevan's Sword or the area with it until yesterday, so I know there could be other obscure quests I've missed.
Also, I'm a little curious what happens to the advisors and the war table, but obviously I'll eventually find out, regardless.

Hmm... as far as I remember, what I would definitely recommend is getting the Forces perks upgrading maximum focus (Advanced & Master Focus).

Nothing matters in Trespasser that you haven't already done.
As for what you should get, the delete button to get rid of that piece of garbage DLC.

There is nothing you have to do before Trespasser its pretty much self contained. Just be happy with your gear and build and I would also highly recommend making a hard save just before starting the DLC for once you start the DLC you cannot go back.

Also make sure that you have upgraded your potions and other things as well.

Hmm... as far as I remember, what I would definitely recommend is getting the Forces perks upgrading maximum focus (Advanced & Master Focus). I hadn't thought of that, but luckily I had already bought them during the main quest. Also make sure that you have upgraded your potions and other things as well. My completionism appears to have covered me pretty well, since I had those already, too. I was mostly confident I had everything crossed off, but wanted to put out feelers to make sure. I'm not too worried about it, but I noticed that all the characters had cut scenes during the initial gathering (which is all I've played of Trespasser so far.) All of them except Iron Bull. Out of curiosity, is that generally the case, or is it likely I might have screwed something up with his approval during the original playthrough?

You may have missed a few conversations scenes with him to trigger his friendship. Or did you let his Merc group die?

Hmm... as far as I remember, what I would definitely recommend is getting the Forces perks upgrading maximum focus (Advanced & Master Focus). Yeah other than making sure you max out your focus there's nothing you absolutely need as the best gear in the entire game you already get in the first 2 DLC's really. As you'll ge tplent yof use out of your focus abilities in this DLC. You'll find out why and how when you play it.

I wouldn't worry about anything besides making sure I have the best armor/weapons I can get and my character is already at his/her strongest.
Just... finish everything you personally care about before starting the DLC.
