I haven't played since launch and am starting new playthrough. I'm looking for important things that are missable or important "paths" that can be screwed up...or not even known about. An example of that is softening Liliena. That is something you can completely miss. I don't need specifics on how to do them, I can google for them if I know what they are.

What world state are you importing and what race/class are you going to play? Do you have all the story DLC and latest patches?
-For Dalish Inquisitors, there is the WT quest line that can get your clan killed if you do it wrong. -The Warden WT quest line if you recruit them after Adamant can get them all killed if you do it wrong. -If you have Alistair as a Warden who did the DR, then if you want him and Morrigan to meet at Skyhold you need to do WEWH before meeting Alistair in Crestwood. Just to make sure I didn't mess it up, I did the ball before even talking to Hawke at Skyhold. -For DLC, you gain a new Mark ability in Jaws of Hakkon that is very useful in Descent.

World state is kinda a big question, it's a custom imported one from the keep that I made as best as I could remember from my previous game choices. Playing an Elven Mage <----So everyone in Ferelden hates me :-) (I'll look into that WT line) I have all DLC, but never played them. I think I made Alistair king. Also...be nice with the abbreviations...I haven't played the game in a over a year. Since you haven't played with any of the DLC yet, you might want to check out this thread:

Trespasser added upgrade toggles for all abilities, so once you upgrade something you can switch between two different versions of it if you want.
Take the recommended levels for the DLCs seriously - the enemies are a lot tougher than in the vanilla game.
Make sure you do everything you want to do before starting Trespasser since everything else will be locked out once you start it.
The last patch added the Golden Nug, which will allow you to sync all your Tier 2 and above schematics, as well as collectibles like potion recipes, seeds, mounts (except the war nugs), banners, etc across all of your characters. To activate it, make sure you are connected to the DA servers, load up the last save of your previous character (if you still have it), then use the Nug located in Undercroft. Once you've done that, every new character you create will have access to everything that was uploaded by syncing with the nug either in Haven or later at Skyhold. As you find new stuff, you can keep syncing so that any future characters have access as well.

Yeah, I found the nug and logged in my old character still sitting on their servers. I also saw the trespasser warning on my old characters WT, it jumps forward 2 years and saw other warning about the DLC being harder.

The Hinterlands is not as bad as its reputation, but it does have difficulty spikes, which makes leaving a practical as well as story-led consideration.

Use some challenges. There is a challenge that scales all enemies to your level if you are higher than them, use this with half exp. Why? The combination of these allows you to do every single side quest in the game without ending so much more powerful than your enemies. This way you can stay in any map you want for however long you want. Its one of the best things that came with Trespasser.

Maybe bit late, but: a) Play with character creation. It is big enough to spend some time here and you have a lot of time-screen with your character (adding this one since friendo always go with one of the default faces and then makes big eyes how beautiful or cute characters someone makes). And if you forget about something (for example, I started to play again and my Inquisitor looked as if she met with shovel few times too much) there is always mirror in Black Emporium (if you have it installed) When you get to specializations always go to tactic screen and put the special skill on use. I didn't do it and wondered why my companions run around with maxed focus (focus? me sorry, me no native english speaker) c) If you have enough of your class and want change BUT you are in middle of gameplay, simply play during fights as one of your companions. I found mages really OP with their range for spells (don't judge me, since I play I've been always rogue only) and I couldn't help myself but play as mage this time. It hurts tho with opening rooms in Haven (under it) and I cry inside. But I'll be stronk! d) As SofaJockey wrote, leave Hinterlands if you have enough of it. What you need from there from beginning is simply go down, kill few people and talk with Mother Giselle. All e) Check them links people posted above me. I found them during my gameplay already when I reached Skyhold and with few I facepalmed a lot that I missed things f) Read perks you can get and think about them. If you want to collect everything, from Secret tree reach perk with opening locks. BUT when you gain Agents they count as well so no need to spend your perk on something you aren't happy with g) I think it's somewhere in the text from link, you can't re-take your perks. Word of advice from me - take all perks with knowledge. Since there is one mage in Hinterlands, I would recommend Secret kownledge perk about magic. Useful, for me h) If you want some nice talk between your chars, don't make same mistake like me and don't wonder around with only Cass, Varric and Solas >.< I found about it already when exploring Hissing Wastes and again got bruises on my face for my stupidity. There are a lot of talks between all of the chars. When you get Cole almost everyone is suspicious about him and would love to kill/throw him away. During gameplay some people can befriend with him, for example Cassandra can be more frieldy towards him, he can tell her stuff about one of the Seekers. There is TONE, if not more, between Cole and Solas. Dorian and Bull. Cole and Blackwall. Bah, there are even talks between all three of your companions. i) Enjoy, play as you want, and... ask people here. I believe everyone will like to answer you or give you some advices. If you want to have from Inq as much as you want, don't rush, go explore! \o/ I've spend over 260 hours and still didn't found one special shield or original chainmal armor And, at the end, word of advice from my playing: j) Explore. 0_0 There is so much hidden. Look at the rocks, at the paintings, buildings, read codex entries... k) It is not really necessary to always have warrior, rogue and mage in team but it would be wise to always go with that team-comp. There are missions where you need only rogue and can take whoever you want (but I do recommend warrior as he is tanking everything *bows to Cass*), there are a lot of Veilfires which requires only mages (so it should be, always take mage). I remember my *disguised noise* when I went on bottom of dungeon and there was wall to break only for warrior. Guess what, climbing on top, going to camp, then long way back to dungeon on bottom and crashing the wall for two chests with... nothing important/stupid stuff to sell only l) Simply enjoy. And use F5 a lot.
