I bought Dragon Age Inquisition on sale but haven't played it yet. Due to a list of games to play and mixed reviews of Inquisition I've read, it wouldn't surprise me if I only played once, rather than the several playthroughs I did of Origins, Mass Effect, and Mass Effect 2.
For those who have played, particularly those who played multiple times, what should I do to maximize enjoyment of that one run?
If you answered this for me on the other board, there's no need to repost since I'll read both. Thanks!

I think it depends on you. I prefer dwarves myself, but there are limited LI if you are into that sort of thing. In DAO mages suck, but they are quite interesting in DAI. I like the rogue in DAI, but have not played a warrior yet.

Having played every race and class, my favorite - Elven Mages. Elves tie in closest to the plot and Mages were the most fun class (for me)!

I can see why female elven mage would be a good choice for a single play though. It would hit most of the major plot points and give the player a larger connection to the over arcing themes of the game. Its hard to explain why without being a massive spoilerwhore.

Personally I felt like most of the dialogue was designed around the idea that you would be playing as a Human, probably because there wasn't going to be multiple races for the PC until the delay happened.
So while I did play as a female Elven Mage, keep in mind there might be bits where your character is expressing confusion at things they ought to know such as Elven lore(without getting into spoilers about what that is). There are still a few good tie in bits with this setup though since as noted, Elven lore does tie into the story a fair bit.
If you're into romances, Female Elf also has the most options.
I haven't done any of the DLC so I can't comment on that.
As far as the content itself goes, I'm gonna say don't worry about hitting up every nook and cranny if you aren't into the collection stuff. One of the more frequent complains about the game is that a lot of the zones are filled with pretty boring side content.
The companion stuff is solid though, at least the actual quests they give you. A few of them are pretty much just approval farms of collecting things, but every one has their own quest as is standard for BioWare games these days.

I would recommend an Elf Mage - KE or RM. I just feel the connection to the lore makes for an insightful experience. I've played as a Qunari Warrior and a Human Rogue as well, both were a blast, so if you're not into the lore stuff, those are good options.

To avoid certain frustrations that come with melee combat due to the game's weird-ass implementation of hitboxes, play a ranged class since your projectiles will just act like homing missiles. Avoid single target melee like rogues, since you can't move during the attack animations and the hitboxes are plain odd. Hint: imagine an invisible human sized target area below every health bar (many creatures have multiple health bars, each is an indicator of where the hitbox is) and you'll have an easier time.
That said, I played a human templar with a focus on two-handed weapons, a drug addicted badass who could chew through demons like nobody's business. Templars can detonate their own primers (like Wrath of Heaven -> Spell Purge), so you can create some combo mayhem without switching characters.
Story wise it is not quite as strong as an elven origin, but it's a solid choice for that entire Herald business.

There's really no answer imo. Go with your own desires and play styles. Do not expect any contentious world effect from racial choices, there isn't much of it. The weave for the DAI tapestry is pretty standard, the events and consequences run the same path for every one.

In terms of lore and story beats, I'd also suggest an Elf Mage. Also, if you bought GOTY edition / similar and want to add to the mindfuckery, roll a female Elf Mage and romance the unique romance option available to that race.
In terms of Narm and lols, go for a Qunari and pick any of the romances available to you - the MoCap is the definition of Unintentionally Hilarious.
Other than that, any choice you make is almost guaranteed to be as good as another. If you care about that, female Dwarfs and female Qunari have less romance options, female Humans and Elves have slightly more. All backgrounds have some unique dialogue / references to their past / special dialogue options specific to that background but it mostly comes to play during the first 10 hours of gameplay.
Class-wise, go according to your playstyle: every class has something really cool going for it.

Honestly any race/gender/class combo works just as well as the next for story purposes. It does feel and look really weird to play as a Qunari rogue due to the disconnect between the combat animations and your frame, especially with a male Qunari. If you're into the whole romance part of the game there is one option in particular that does stand out for being more tied to the overall story than the rest, and it's only available to female elves.

For me - I loved my first character - a dual daggers female Qunari rogue. Felt awesome and punchy.
As others have said, female Elven mage (my current playthrough) has a lot to offer in terms of 'tiny extra tidbits' + the whole Solas thing.
Mages are also a lot more fun than in Origins.

After a few false starts I finished the game with a female elf archer. Reasons:
- Alix Wilton Regan. - Melee weapons are odd in this game. I kept unintentionally hitting the air. - No need for Sera.

Like a few others said - Female Elf Mage (Solas romance)
There are some very major plot points that will hit a lot harder for that character, that will just be kinda interesting for others. The other characters you could play don't really have anything special and unique as an alternative. The only downside is that there are 1 or 2 lines of dialogue that don't quite fit - you can actually avoid most of them by not clicking on them. I'll take extra plot relevance over avoiding a few awkward lines.
There's also the fact that there's not really another major character that fits into that mold in the game. There's no other major Elf character that fits the typical Elfy viewpoint, so it fills in that gap.
(The mage part is less important than the others, but it does get you some extra lines throughout the game and there's an extra Elfy spec you can take to really play a full Elfy character)

I'd add that it's a good idea to set up your choices from previous games in the Keep. The stuff you did in DAO and DA2 doesn't have a huge impact most of the time, but there are a few nods and cameos that make your playthrough more interesting.
Other than that, I agree with the female Elf mage thing, but only if you're into the romance content. Otherwise the game does seem more oriented towards a human protag. Get out of the Hinterlands early and don't feel pressured to explore all the zones in the game (I missed 2 of them entirely on my first playthrough.) It's worth doing the main optional questline in Crestwood. IMO it's one of the best side quests in the game. Make sure you watch the after-credits scene, and consider picking up the Tresspasser DLC specifically - I think it improves upon the game massively. Most of the rest is just up to your discretion as a player. Do what seems the most fun to you and don't get bogged down with fetch questing and resource gathering!

I suggest female elf. It gives you the best opportunity to get the most emotional bang for your buck...she also has the most romance options (6). After my own first PT as a female elf, I advised my husband to play a female elf. He has since played as a male human rogue. He much preferred the elf PT. He's usually not big on game romances, but he got really invested in the romance which is restricted to female elves only (as did I).
Having watched my husband's human PT, I really don't see how it makes more sense plotwise. People who say that may be more into the Andrastian religious aspect of Dragon Age. A fair portion of the base game and the DLC also feels more relevant to an elf, male or female.
Other random tips: I second the tip to use the Keep before you play and to leave the Hinterlands early. Be sure to talk to all your companions and do their companion quests, or you'll end up missing out on some interesting stuff. You won't hear companion banter if you use a mount, but on huge maps mounts come in handy. Be sure to dismount from time to time so you can listen to the banter. Don't bother with all the fetch quest or "requisitions," but be sure you explore. My favorite area in the game is only available after finding an item tucked away on another map. You can craft really good equipment, so don't worry about saving up a bunch of gold for prefab armor or weapons. Buying schematics is more economic, as you can use most of the schematics to make stuff for all your party members.
Of the classes I've played the rift mage and tempest dagger rogue were the most fun. You could always play a little MP to see which class is your style.

I have played Inquisition 9 times now but if i would give you an advice then go with Elf Rift Mage that was for me the most fun protagonist.

I never use mods on a first playthrough, but if you happen to play on PC and are only playing once consider installing ConversationCameraZoom. It has some framing issues, especially with qunari and dwarves, but it brings the camera in close when speaking to NPCs and makes the game feel more personal (plus you get to actually see how nice the characters look). After using it a short time it'll be easy to get great angles in most conversations.
