Have a feeling Purple Hawke would win by a landslide... Probably for the best they aren't in this.
It’s difficult not to ‘just’ pick the character you like irregardless of their funny moments. Also I agree, I do not remember half of these characters.

I do wonder if you mean deliberately or unintentionally funny, by which I mean they can come out with something intended even by the writers in all seriousness but actually makes me laugh. Anyway, here goes:
Group A: Warden's Mabari - that dog can always be guaranteed to lighten the mood!
Group B: Sten - it's the deadpan delivery that does it!
Group C: Wynne - mostly as a stooge for Zevran!
Group D: Sister Theohild - come on she has to be doing it intentionally, everyone else takes the Chant of Light so seriously!
Group E: Zevran - life became so much more bearable when he joined the team.
Group F: Shale - it might have been Teagan but not after Trespasser
Group G: Alistair - once he stopped moping about Duncan.
Group H: Stupid Bridge Bandit - could end up being the dark horse in this race.
Group I: Chicken escaping Shale - did she actually miss any or is it the fact that it didn't?

What? No Herren and Wade to chose?!? What is wrong with you?  Sure nothing beats Purple Hawke but Herren and Wade are way more funny than the supposed comic relief Sandal (and Bodahn?) Honorable mentions: Merrill, Aveline, Esmeral Abernache, Movran the Under, Vicomte sorry Provisional Viscount Bran Cavin, Carroll, Baron Arlange, Fergus Cousland, Mad Hermit, Dulci de Launcet (her son Emile is more of a sad than a funny character), Felsi, Dagna, "I am not a mage" Dalish, Yvette Montilyet, Talkative Man, Zither, Your Trainer, Javaris Tintop, Hubert Bartiere Old Tegrin (“There's two things I'm here for –travel or trade. Understand?”) and funniest character of them all Smith.  |

Anyway, here goes: Both! Anything that made you have a hearty guffaw. For the Stupid Bridge Bandit, I always found it funny that if you intimidate them with 'do you want to fight a mage?' line, he shouts, 'Heeeelp! I don't want to be a toad!' and runs away. Obviously laughing at him there. 
As per Chicken, I've only seen a cut scene where Shale didn't miss. I just thought a random image of some chicken getting their feather ruffled to get out of the way was funny and made competition for the baddie bracket. Have a feeling Purple Hawke would win by a landslide... Probably for the best they aren't in this. Oh, I don't know. I found DAO Warden's lines to be equally funny. 
I've been debating whether to add Hawke/Warden/Inky to the poll, but then we're sort of supposed to be the tuffest, wisest, snarkiest type of thing. Still not sure if we want our heroes on the poll too.
I'm doing DAO poll. But I'll do DA2 next.

Bodhan is on the list in Group F. I guess I should have added the as a pair too with Sandal.
(This one is though between Zevran and Flemeth)
I found that group to be the hardest one hence Daveth is funny too. When Jory is chickening out in the Wilds about Darkspawn ambushing them, Alistair says he can sense darkspawn, so no ambush. And Daveth is like, 'See Sir Knight, not to worry. Darkspawn will get us, but we'll be warned about it.'
Alistair - once he stopped moping about Duncan.
*gasp* What about sassing the mages and naming a child after one of them? The grumpy one.
