Depends on how you spec your passives. For me it is Singularity. Because: 1. Enemies affected by Singularity (or any Biotics) take 20% more damage. 2. When one of them are affected by Singularity (or any Biotics), My barrier increases by 40%. 3. I am a Vanguard So tis Biotics + Combat. No Tech!

Pick 2 should always be the special ability IMO, so Charge, Barricade, Remnant VI etc.
On PC I swapped the controls around even to simulate the intended feel so that ability 2 was on C and Mouse 4 and 5 were ability 1 and 2.
All my builds thus far use 1. Overload 2. Ability of choice 3. Energy Drain
1 and 3 trigger a tech combo and does wonders on the many shielded super-enemies that kill you in 3 seconds on Hardcore and Energy Drain gives me 50% shields back, so that's a survival technique I learned was necessary from the get-go. Then sometimes I add in Charge in the middle so I can fully replenish my Shield in an emergency.

WHy not pull?
You can also pull enemies towards you and subsequently nova them.
pull also damages shields at a final evolution.

Also with Singularity you can sometimes get off two biotic explosions if you use lance immediately after charging.

I went with Singularity and haven't regretted it.
AoE CC, high damage even on insanity with right spec, ability to prime any and all enemies in the game, +20% damage against anyone effected by it, long range, auto tracks targets for nearly 100% hit rate, hits enemies through cover and wall, long enough duration where you can get several primes and hence combos from one cast, can be detonated early for heavy burst damage if spec'd for explosion... You can use it to plug a choke point, you can toss it at your feet and it will protect you from any CC'able melee mob.
There is literally no situation where a fully leveled singularity isn't awesome.
IMO, singularity is the second most powerful skill when fully leveled. The most powerful being charge.
IMO, the only time this should even be a debate is in the very early game when you don't have enough skill points to get to tier 6 on the skill.
