Ships on FTL are undetectable. Once they jump, they are safe. But their destination can be calculated from the route they take . Once the Reapers find it they would simply need to jump there and wait since they move faster at FTL. So I believe it's unlikely the game would start during the Reaper War. Anyway, i think it will start inside the Arc Ship with the protagonist not knowing if he/she reached Andromeda.

It'll start with the opening movie showing the Ark's, their preparation and leaving earth, It'll show you in a cryostasis pod which begins the character creation setup. You then wake up where the story begins...

What I'd like to see and what I think we will actually see are oceans apart.
I'd be happy with a cgi opening scene with the different species getting on shuttles and lifting off from their planets and flying to meet the Ark ships with a voice over explaining how the galaxy is under serious threat, a threat we are not sure we can beat, and that under the guise of an exploration mission (so as to not cause mass panic) all the galaxies governments had agreed in secret to send enough people to ensure their species survival. *images of mixture of races loading onto the Ark and into pods*
Then something along the lines of the Arks firing up into FTL, leaving us staring at empty space, then the voice over saying 'we left just in time' then just the trumpety blargh noise the reapers make, then cut to ME:A logo.
Then a tracking shot of pods until get come to Ryder then CC then 'we arrived' scene.

Which they would if they thought they might be being tracked

Ehhhh, kind'a but not really. Those kind'a distances are so hard to tell by the angle of the ship in 360 space, simply because of .0001 difference can mean about 5-10 star systems. And that's on one axis. Take into consideration that there are three axis -- Then multiply them all by each other, you're all of a sudden looking at 30 to god knows the calculations... I know it's high though.

Nothing prevents a ship from changing it's course. And when you see this ship it's no longer there. Every minute it passes 0.2 trillion kilometers, one degree deflection gives millions kilometers window. It's worse than trying to catch a fly on football field.

The ARKs are not using standard FTL drives because they arrive in Andromeda in 600 years. So, all the restrictions like discharge cannot be applied. The only way to avoid any existing canonical restrictions is to re-write the rules or create a "FTL discovery". Here is where the prequel novel comes in to re-write those canon texts. I don't see the game explaining the canonical changes. That is not the function or purpose of the game.

That would require leave FLT to plot a new course correction, but it's true the search area would be colossal.
However the Reapers could still use this to estimate where the Arc Ship would be as long as the destiny (Andromeda Galaxy) remains the same. And the Reapers are very methodical, remember they are the beings who hit planets from orbit until they lost all liquid water and atmohspere to space just to wipe past civilizations.
edit: I believe that is too much especulation Sartoz. At least until the books/comics etc give us something about it (if they do).

It starts right out the gate with you surrounded by the enemy. As you fight them, more and more show up. Just as you're about to attack, everything goes dark and a critcal mission failure screen shows up.
The character creator pops up. After which Ryder turns around and yells at the person who caused the death of the player in the video game. The guy apologizes, but tells Ryder that its nearing time for the ships to leave the Milky Way.
A cutscene shows shuttles approaching the ships. The scene switches back to Ryder who gives one last hug to family members before getting in his/her pod.
The ships are seen entering ftl. The title Mass Effect Andromeda shows up

Maybe the game starts with someone (a relative who is part of the Project) revealing the existence of the Project to us. Why? They need Ryder for a specific reason (scientific knowledge, protection of important scientist, discovering who is a Cerberus mole, etc.) before it is too late. You start playing five minutes of introduction before something (Reapers, Cerberus, indoctrinated people, Leviathan, X) starts to attack the Project.

Well, a lot of it will have to do with who the enemies are in the MP. If the reapers are introduced, they will be in the MP. So, I'm guessing that we are fighting off Cerberus. Another reason for my theory of starting from Sanctuary.

The main menu will show the room with all the cryostasis pods as the ARK glides silently through space.
Pressing the start button will initiate the wake-up protocol.
Camera zooms in to Ryder's stasis pod, lets you pick a first name on the pod's name plaque, then moves to show Ryder's face.
Character generator starts.
When you are done, Ryder wakes up. "We made it."
Text popup - new codex entry: "The Milky Way"

I don't think we will see the Milky Way at all ( and I personally prefer if we don't... )
My bets are on seeing the Hyperion floating in the space, then the camera goes inside showing different deactivated levels, all silent, and then how the ship starts activating them. More zoom in and character creator shows up.
The information we have so far makes me think that we won't be thrown into action right away, that would mean that as soon as we enter Andromeda we run into a space-faring species that doesn't like us arriving there... unless the "action" comes from the ship itself, who knows, may be something breaks and cannot be repaired by automated means. I discard conflict between the passengers since everybody is supposed to be in stasis...

We don't wanna forget that "600" was considered a good guess, not the actual time. It's just as likely, taking the Black Ark theory into consideration, that it'll take less time. Even if the theory isn't used, they're bound to have found something from the Collector's which'll be similar in tech from when the group Vega was in gained Collector data during ME2's timeframe.

I'm kind of hoping it starts with some sort of mechanical glitch that Ryder has to fix, which is our how to move tutorial, followed by a return to stasis. Just something fairly sedate and picking up in the middle of something bigger, with discussion after stasis.
Given that it's Mass Effect, I'm hoping things will be absolutely terrible when the titlecard plays. (My reaction to the opening of Mass Effect 2 was "Good game. A little short.")

If you think about a process where you go to sleep and then 600 years later and you wake up, but for you, no time has passed, the last thing you'd think before you go to sleep is, "Are we going to make it? Am I going to survive this? Will the ship just implode somewhere along the way? So we actually wanted to capture that moment of, "Oh my god, we're here!" That's really where this story begins.

In any case, the studio did say that it's not the purpose of the game to flood the player with a backstory. If you want more, the prequel book Initiation is will be available.... I'm thinking Nov-Dec 2016.
