It might be a bit far-fetched, but here we go.
I was watching the vid unveiling that Natalie Dormer is Dr. Lexi's VA and one of her lines called my attention. It goes something like "It's a seizure, her brain can't handle the connection!"
So what if that's the reason why Cora doesn't step in as the new Pathfinder once something happens to DadRyder? Her brain cannot "connect" to the ship for whatever reason and they default to you because of Alec's mystery project.
Dunno, maybe it's a bit too obvious and everybody already figured it out, while I'm here thinking I'm grasping at straws.

It makes sense to me.
The first time I saw the video, I was thinking that line might refer to Sara Ryder if the player is Scott, but Cora works too.

Because Alec Ryder is a nepotistic dick, that's why. No, really, I am rolling with theory that Alec is kind of a dick until proven correct or wrong when game is out. If he was young and inexperienced, I would consider him to be a dick because he's just hard for no reason. Considering Alec is older, wiser, and been through a good bit of grizzled combat, I'd say he's just an old man molded to be cold and commanding while stuck in his ways due to his previous encounters with hostile aliens while traveling to the unknown. He's a product of his environment and past as well are. Except dicks. Their just dicks for no reason at all.

Very suspicious that Miss Cora Harper possibly related to Jack Harper, hero of Shanxi, and second to Alec Ryder, N7 hero of Shanxi, was incompatible with that SAM implant that shares a similar origin (Hannibal VI) with that Cerberus AI EDI. (cough)

So it is probably due to deep and long physical connection with Ryder family that causes it.

Don't know if it should be put in spoiler but if we say that only the Ryder family can be connected with SAM in that way it is shown: There is the fact that Ryder can interact with the remnant tech which makes us "special" for the Archon. It may be because of SAM and that put us in charge. Anyway,it will be fun to see how much we can tease Cora about not being in charge. Or if she is going to act as "brakes" when we want to do something stupid. If her personality is as they have mentioned it should be logical that she is going to act more mature and try to make us avoid doing stupid things.

Cora being higher in grade then both siblings and the designer successor confirmed that.

I think no on nepotism because of the potential for Cora to romance one of the Ryder twins. This would create a rift between them, I'd think. The inability to connect with SAM might make more sense.

It happened with Miranda, too, but for slightly different reasons. I imangine Cora will be jilted and hurt if DadRyder chooses the plucky newcomer over the Askari trained badass. But it might have something to do with the way Alec gets ousted—if he has a stroke or even gets a concussion in the line of duty it would wreak all kinds of havoc on the SAM connection. But since Scott and Sarah are Alec's kids their nervous system could be similar enough to shoulder the transfer of SAM's control.

I also think it's good old "boss said so"... the Ai is not really a military operation, so rank isn't a factor for promotion/succession, and Alec Ryder is the commander of the human PF team.
I think the seizure line is actually referring to the non-player sibling, since they need a reason for him/her to not be part of the actual away team, and all other squaddies are equipped with SAM implants.
