I'm going to have to jump onto the Vigil/Ilos bandwagon. That whole scene was a masterpiece of plot exposition (honestly, Bioware have only equalled it with the final conversation of Dragon Age Inquisition: Trespasser) I think both the conversation with Vigil on Ilos and the earlier exchange with Sovereign on Virmire set the stage for the entire trilogy.

Thanks to the OP for starting this - it's a less problematic topic for this forum and its inhabitants I also started ME2 first back when DA2 was first released and Bioware gave the Deluxe Edition owners ME2 as a gift. I loved just about everything about ME2 including the dramatic opening sequence - what a way to start a game. But the best was the entire sequence of going through the Omega 4 relay and the suicide mission. In ME3 (with all DLCs) it was the opening leaving earth sequence just reeled me in and had me rivetted until the very end which was equally emotional. It was awesome to see the majestic Normandy arrive for pick-up. It felt like home. Unlike many people, I actually enjoyed the ending although I would have loved to have seen my femShep and Garrus live out their retirement from vid royalties Awesome games and too many moments to choose from. Edit: the only moment from Andromeda for me is the wtfbbq braking of the Tempest as it hurtled toward that huge Kett Archon ship

But the best thing for me really is, that my playtroughs are still after all these years different and I manage to find still new things. I did not know I had my own mission based on my background! (I usually play as an orphan raised on the streets since I had a difficult times as a kid.)

Andromeda wise for me was that great intro. The slowly building music, Clancy Browns epic voice, the shuttles skimming the moon and arriving at the Arks just as the music and the traditional ME wordy bit kicks in - amazing! Then followed by the Hyperion arriving at Heleus with Day 0 Hour 0 at the bottom of the screen. That really made me smile!

I don't think I could pinpoint one specific 'moment' for me, but one side mission did encapsulate everything that made me fall in love w/ the OT. I Remember Me. It's only available if Shepard has the colony background and it includes a number of dialogue choices and actions that defines the character of Shepard.

My son (about two years old at the time) watching Mordin sing on the ship. He was laughing so much and kept asking me to replay it. It was heartbreaking remembering that with what happens in ME3 to Mordin.

I think my favourite moment was meeting Sovereign for the first time and just how much that scene raised the stakes of the entire mission. It's one of my favourite villain moments in any form of media. The fact that Sovereign doesn't fall into the trap of revealing it's plan, but still delivers a haunting speech about millions of years of genocide, just makes it even better.

I'm enjoying reading the reminiscences - I couldn't pick a favourite moment I think though I will say the Vigil meeting was amazing. For one thing, Vigil's theme, which I love beyond all the ME music. Then, when I played that scene the first time, I remember getting this eerie feeling creeping up my spine: a voice of a lost people just audible to Shepard over an immensity of time - that was thrilling.
The other big thing for me was realizing you could play a female character. I came to ME a bit late, recommended to it by someone on a forum somewhere, and thought 'oh space thing - it'll be all a macho boys thing'. I loved femShep.

It wasn't really hard to get me hooked since I am a avid fan of sci-fi and of video games. It was a done deal the moment I stepped foot on Eden Prime for the first time. There was absolutely no doubt after Virmire. I had such a hard time with the choice, not because I really liked both characters but because I left Kaidan with the bomb and I was romancing Ash. I had to protect the bomb! This is one of the reasons why this was also the only mission (until ME3 came out)that Kaidan survived in all of my PTs which there were countless PTs.

Love at first sight.
For me it was from the very beginning. It was a big step up from Jade Empire in every aspect.
I chose the bottom line in the first conversation with Joker and Femshep said, "Great. You pissed the Captain off, and now I'm gonna pay for it". The delivery of the line was very real, not like a cartoon voiceover, and Shepard side-eyed Joker as she left. I noticed the realism of the scene. Then there was the chat with Pressly. Femshep asks, "you think the Alliance brass is holding out on us?". Again the facial expression added to the line to produce a great cinematic effect. By that time (2 minutes in) I was absolutely drawn in. What made it was how alive everything was.
I had not experienced that in games. Alyx Vance from Half Life 2 was closest but still not as good.

I honestly have no idea. Really I don't, there are so many moments... But if I have to pick one, the very first time I thought this could be something special was when the turian spectre was assassinated by Saren.

Then I felt a little odd standing in a giant mushroom world... it felt more like a fantasy environment than sci-fi I used to love in ME1 and ME3. Then came my fathers death and the disappointment started to grow too much... EA has nothing to do with MEA:s loss. It was the lead writing team that was unfocused and let too many opportunityes slide off their hands. Sorry about the OT.
