Walking around the Presidium in ME1 was my favorite moment. It was a huge, hand-crafted space station with aliens everywhere. It was big enough to actually get lost in it, which I liked because if you're not getting lost then how big could it possibly be? It set the tone for this new world you would be immersed in with all it's different characters from the quirky Conrad Verner to the cold Nasana Dantius. I would've liked for there to be more stuff to do there because some of the walkways were pretty long and there were some walls that could've been used for other things but I thought it was cool anyway.
The conflicts were great and really different from most games because the combat areas didn't give themselves away with chest-high walls. In fact, firefights could just happen out of the blue in the wards and you never expected it. The wards were said to be dangerous but I never actually believed it, thinking it was just random lore but people really did tend to shoot at you down there. Even conversations could end in firefights. So you never really knew what to expect while you were there which is a really great thing that you don't get from a lot of games.

Despite many, many, many awesome moments in ME games, the moment of complete rapture for me was when I was informed on the forums after recruiting Thane that he can be romanced. It was literally the first time ever that a char I wanted to romance was available.
i know how silly that is, and how peeps look down on romances, but, well, I got my moment there, and now I can start not caring and not consider it essential.

Since this question is rather loaded towardds OT responses, I'm going to treat ME:A as a new story and give my moment with it:
"Is that our golden world?"
The second moment was when Jaal realizes who the Kett really are (I did this quest shortly after completing The Lost Scout... where you come acrosss the Angaran saboteur torturing the Kett in order to find out where her family had been taken. My Ryder opted to shoot the Kett and insist she return to camp; but as Jaal ask "How many have I killed, not knowing?"... it hit me that, perhaps, I had killed the very member of her family for whom she had been searching.)

Lets see. ME moments, eh?
ME1: I think this one has the most moments for me. Approaching the Citadel for the first time; the conversation with Sovereign, shooting out the elevator on the Council Tower climb, the whole of Ilos (especially Vigil).
ME2: The first time I charged as a Vanguard and shotgunned an enemy to the face. Discovering who the Collectors really were. Lair of the Shadow Broker DLC (the whole thing). Oh, and just for fun, the first time I sniped off the head on those big mechs.
ME3: Mordin on Tuchanka. Shooting bottles with Garrus. The Citadel DLC. Sometimes I'll just go back and play that because it's so fun.
MEA: DadRyder's speech on the shuttle before Habitat 7. Going through a Vault for the first time and the escape. The whole "Benefactor" and the Ryder family secrets arc.

ME1. The first mission (tutorial). It was a good introduction to the series.
ME2. The Suicide Mission. Its success or failure depended on the previous choices made, strategical and tactical decisions.
ME3. Walking and shooting Cerberus troops with Black Widow (without stopping or looking for cover) on Tuchanka, when one bullet equaled one corpse; Jumping into the green ray.
ME:A. Escaping from the purification field at the first Remnant Vault.

I don't remember the name of this place (a room within a strip club, I think) so I am including a screenshot for reference. This whole area from recruiting Wrex to the strip club and shooting the place up was it for me. I was a big fan of "Duke Nukem" and selecting the red renegade lines for Shepard just tapped into that vibe. Being able to play like that as a female character sealed the deal.

the first time I sniped off the head on those big mechs. Yeah that was very cool unless you were like me and were standing close enough to be caught in the blast. It didn't take me long to experience the mission failure screen in ME2.

Legion was such a great moment because I was so surprised that a geth was the last mate. And then he turned out to be one of my favorite characters. Plus the mission to reach him was the hardest for me to stay alive in because of all the husks and Scions, so finally getting to him felt really felt good...

Sorry to continue from this, but after the great opening everything started to fell apart. First thing I noticed was the new discussion system. Then another disappointment came when I realized that my brother is unconsious and I won't be contacting him anytime soon. Then I felt a little odd standing in a giant mushroom world... it felt more like a fantasy environment than sci-fi I used to love in ME1 and ME3. Then came my fathers death and the disappointment started to grow too much... EA has nothing to do with MEA:s loss. It was the lead writing team that was unfocused and let too many opportunityes slide off their hands. Sorry about the OT. We get that but what is your best moment in mass effect? I already told it in my earlier post :-). I love the whole Noveria mission. The port Hanshan has a special place in my heart. I love the architecture and the small missions. I have to say also that turians are just goddamn great.

I came in... Late.
Maybe a year after ME3 release, with all the DLC out. Inquisition was my first BioWare release, and Andromeda was my first Mass Effect release.
To be honest, I don't know how I came to it, but I tried the demo for ME2 and ME3 and thought RPGs were cool as fuck - I haven't actually played that many before, which is hilarious. RPGs became my favorite genre afterwards. I just... Knew. It clicked with me on a lot of levels. So I got the Trilogy... And eventually all the story DLCs, together with a few weapon and appearance packs. One day, I'll actually finish the collection.
But when I played the Trilogy for the first time, the moment that made the entire journey, the one that really shone the exceptional first impression that I had not been wrong, and in fact been GOD'AMN RIGHT was jumping through the Charon relay. Began there, and certainly did not end. I've pulled a few all-nighters on my PC before, but NEVER was I just so deep in a story to just lose track of time. No, every time before the Trilogy, I knew precisely what I was doing. In ME3, with the Rannoch arc was the first time I just looked away from a story I fell in love with and realized it was dawn outside.
When Andromeda came, I almost gave in to the skepticism surrounding early. That would've been a mistake. The menu music in the Trial was as beautiful as they came, and then there was the New Game. We landed on Habitat 7, and what was supposed to be a time of exploration went downhill faster than you can say Keelah se'lai. When we met Alec Ryder, he commanded me, Cora and Liam to attack what we thought was an atmosphere processor. He tore through the enemies effortlessly. That N7 did not need our assistance, and when we fought our way to an alien console, he gave us the reponsibility of defending our position as he and SAM gained access. When he interfaced with the inner console of the processor... That is when Andromeda was made for me. It clicked, like the Trilogy before, and a new era began...

Walking around the Presidium in ME1 was my favorite moment. Great shout there danishgambit, I can still recall the joys of exploring the Presidium for the very first time. Man, that first game delivered so many indelible moments it's really difficult to single out just one. I guess mine would have to be that conversation with Sovereign. Just a legitimately game changing, wonderfully memorable, moment!

There are a lot of moments sprinkled into the trilogy and Andromeda. There isn't a definitive moment for me.
I guess the best moment for me is ending the Morning War on Rannoch. Legion sacrificed himself to add his own personality to the consensus and Tali is standing on her own world. I didn't have a romantic interest in Tali, but it was at the moment that I felt hope on how the final battle will turn out. That same moment was a big tease too! I hear the seals on Tali's mask coming undone and I'm ready for the reveal... still waiting... F*** you Casey.

I agree with you it was great to see them latter on down the road and it was really sweet getting some payback on Harkin. I don't remember exactly what he says to FemShep in ME1 but if it had been me I would have pistol wiped that sob.

Shooting Mordin in the back and tricking the krogan into thinking I cured the genophage.
Shooting Udina. I had been longing for that opportunity since I laid eyes on him in ME1.
For Andromeda, finding out about the mysterious "backers". So many questions left unanswered.
